Feliciano Miguel Rosendo da Silva, 57, was arrested in March 2014 and has been accused of holding orgies at the Order and Mandate of St Michael the Archangel in Galicica in northwest Spain.
Two nuns residing at the convent told police that he had sex with them on a daily basis in order to 'purify' them.
Two women, Marta Paz and Sandra L, were first arrested on suspicion of engaging in abuse, but were later released without charge.
The pair have taken virginity test in an attempt to try and argue the allegations against Mr Rosendo have no evidence behind them.
The Madrid College of Physicians carried out the tests.
Police have not said there is any physical evidence to support the claims of abuse against Mr Rosendo.
He denies any wrong doing and is awaiting trial on charges of sexual abuse and tax fraud.
The Bishop of Tui-Vigo, Luis Quinteiro Fiuza, has suspended the order until the outcome is heard.
