Pastors Joy and John Torrens wrote on Durban Christian Centre's Facebook page: "Our Christian faith teaches us that all things are possible and no matter how tough the obstacles we shall overcome them."
Durban Christian Centre housed around 5,500 people at the Jesus Dome and it took firefighters five hours to put out the blaze.
No one was injured in the fire and the cause is still unknown.
The fire took place on Tuesday night and police spokesperson Lieutenant Nqobile Gwala said they responded to the call of an explosion.
Reports suggest that police forensic teams will look at the buildings CCTV and phone records to determine the cause of the fire.
"At this stage we cannot say what was the cause of the explosion and fire as the emergency teams are still battling the blaze," they told IOL news.
On social media the Durban Christian Centre Jesus Dome says it is "a House of Prayer for all nations."
It also describes itself as "a place where miracles happen. A family you can belong to. Big enough to reach you, but small enough to care!"
Jesus Dome was completed in 1999 after two years of construction. The auditorium features big screens and a stage.
Worshippers are meeting elsewhere for the time being.
Durban Christian Centre Jesus Dome wrote on its Facebook page: "We stand united in our faith to rise triumphantly from the ashes and see the Jesus Dome rebuilt."
