Hundreds of people packed inside the Fleet Street church to celebrate the couple's wedding - a day after they officially tied the knot in London.
The service started at 11am on Saturday at St Bride's Church, which proclaims itself as "the Journalists' Church" offering "a spiritual home to all who work in the media".
Ten children from the couple's previous marriages were among the guests.
The stunning sanctuary promises visitors that: "Whether journalist, tourist, City resident or worker, you will always be made very welcome."
Flanked by his sons, Mr Murdoch paused briefly for photographs before entering through the church gates.
Shortly after, top media figures including Alan Yentob, Rebekah Brooks and editor of the Daily Mail Paul Dacre arrived.
Barry Humphries, the Australian comedian better known as Dame Edna Everage, joked: "I'm not getting married" as reporters asked him how he was feeling.
He said: "I'm very happy. I think it's great, I like them both.
"The wedding present was hard to work out.
"I always liked Jerry but unfortunately I'm married," he joked, before entering the church.
Bob Geldof and the Turner prize-nominated artist Tracey Emin were also on the guest list.
The service was led by Revd Canon Dr Alison Joyce, Rector of St Bride's.
Catholic priest Fr Jonathan Morris, a commentator on Mr Murdoch's Fox News Channel, assisted.
